Rybelsus is a medication based on a molecule structurally similar to the human hormone GLP-1. In other words, it’s a synthetic derivative of this hormone, capable of mimicking its natural functions in the body, placing it in the drug class known as “Incretin Mimetics”. GLP-1 is a hormone secreted in the digestive tract following meals.

One of its roles in the body is to stimulate insulin secretion from pancreatic beta cells, thereby helping to regulate blood sugar levels. Additionally, this hormone enhances feelings of fullness after eating. As for Rybelsus specifically, the medication appears to aid weight loss by influencing areas of the brain that slow stomach emptying after meals and significantly increase sensations of satiety.

Although Rybelsus is not officially approved as a weight-loss medication, many patients who previously took it for diabetes management also experienced weight loss. For instance, studies have shown that patients taking Rybelsus (active ingredient: semaglutide) at a dosage of 3.7 mg lost an average of 2.3 kg. In contrast, patients on Rybelsus at a higher dosage of 14 mg lost approximately 3.7 kg.

However, compared to these earlier studies, a recent clinical trial highlighted the potential of the active ingredient in Rybelsus (semaglutide) to help individuals with obesity reduce their body weight by an impressive 10-15%. These significant weight reductions could potentially lower the risk of developing severe chronic diseases.

The trial included 1,961 adults with a Body Mass Index (BMI) of 30 or higher, or a BMI of 27 with at least one obesity-related risk factor. None of the participants had diabetes, though some were diagnosed with prediabetes at the start of the study. All participants received dietary counseling and a recommendation to engage in 150 minutes of physical activity per week.

Participants who took a higher dosage of semaglutide (the active ingredient in Rybelsus), administered weekly at 2.4 mg via injection, in combination with dietary modifications and exercise, lost approximately 15 kg by the end of 68 weeks. In comparison, those who received a placebo lost only around 2.6 kg.

The group taking the medication also experienced significantly greater improvements in obesity-related health conditions and reductions in waist circumference. Safety profiles were excellent, and side effects in both groups were mild and comparable to the placebo group.

Furthermore, a meta-analysis conducted in January 2021 reviewed several recent clinical studies comparing the weight-loss efficacy of semaglutide (the active ingredient in Rybelsus) to other weight-loss medications. To specifically isolate the effect of each medication on weight loss, researchers subtracted the weight loss observed in placebo groups from that seen in participants who took the medication.

The findings revealed that the medication Qsymia resulted in an average weight loss of 6.8% of body weight; Saxenda led to an average reduction of 5.4%; Contrave contributed to a 4% average weight loss, and Xenical resulted in a 2.9% reduction on average. In comparison, patients taking semaglutide (the active ingredient in Rybelsus, Wegovy, and Ozempic) lost an impressive 12.4% of their body weight on average.
